Detection Principle

Flow cytometers use laser light to illuminate individual cells, generating forward scatter (FSC), side scatter (SSC), and fluorescence signals. Forward scatter reflects cell size (with a resolution of 0.2 μm to 0.5 μm), side scatter indicates the complexity of the cell's internal structure (such as the number of granules), and the fluorescence signal is used to detect molecular markers (such as antigens or nucleic acid content). 

Function of the Optical System

The optical system includes a laser, lens assembly, and detector, which generate light signals and convert them into electrical signals for analysis. Lasers of different wavelengths can excite different fluorescent markers, enabling multi-parameter detection. ‌

 

Signal processing

The detected light signals are processed by a computer to generate histograms, dot plots, and other data for analyzing cell population characteristics or performing sorting. For example, combining FSC and SSC can preliminarily distinguish cell types, and then fluorescent signals can be used for precise molecular-level analysis.
